Na za\u010diatku je pr\u00edtomn\u00fd mal\u00fd n\u00e1dor, ktor\u00fd sa d\u00e1 zisti\u0165 fyzicky, vo forme hr\u010dky alebo sa d\u00e1 diagnostikova\u0165 pomocou r\u00f6ntgenu.<\/p>\n\n\n\n<p>Ale nie v\u0161etky hr\u010dky v prsn\u00edku s\u00fa n\u00e1chyln\u00e9 na rakovinu, preto\u017ee v\u00e4\u010d\u0161ina z nich je ben\u00edgna. Nerakovinov\u00fd n\u00e1dor je len abnorm\u00e1lny rast, ktor\u00fd sa neprejavuje mimo prsn\u00edka a rozhodne nie je v\u00f4bec \u017eivot ohrozuj\u00faci. Vaje\u010dn\u00edky sa tie\u017e lie\u010dia liekmi alebo chirurgick\u00fdm z\u00e1krokom, aby sa im zabr\u00e1nilo produkova\u0165 horm\u00f3ny. Na \u00fapln\u00e9 odstr\u00e1nenie rakovinov\u00fdch buniek sa odpor\u00fa\u010da komplexn\u00fd re\u017eim.<\/p>\n\n\n\n<p>Tu je d\u00f4le\u017eit\u00e9 spomen\u00fa\u0165, \u017ee v\u0161etky vy\u0161\u0161ie uveden\u00e9 lie\u010dby maj\u00fa z\u00e1va\u017en\u00e9 ved\u013eaj\u0161ie \u00fa\u010dinky, ktor\u00e9 m\u00f4\u017eu ovplyvni\u0165 aj in\u00e9 org\u00e1ny tela. V tomto pr\u00edpade je v\u010del\u00ed jed jednou z najvhodnej\u0161\u00edch mo\u017enost\u00ed lie\u010dby rakovinov\u00fdch buniek bez po\u0161kodenia zdrav\u00fdch buniek.<\/p>\n\n\n\n<p><strong>Protirakovinov\u00fd \u00fa\u010dinok v\u010delieho jedu na \u013eudsk\u00e9 bunky rakoviny prsn\u00edka MDA-MB-231 pomocou Ramanovej spektroskopie<\/strong><\/p>\n\n\n\n<p><strong>Konspekt<\/strong><br>V tejto klinickej \u0161t\u00fadii sa sk\u00famal apoptotick\u00fd \u00fa\u010dinok BV (v\u010del\u00ed jed) na \u013eudsk\u00e9 bunky rakoviny prsn\u00edka prostredn\u00edctvom anal\u00fdzy hlavn\u00fdch zlo\u017eiek (PCA) a Ramanovej spektroskopie. Ak\u00fdko\u013evek druh biochemick\u00fdch zmien v rakovinov\u00fdch bunk\u00e1ch bol hodnoten\u00fd po lie\u010dbe v\u010del\u00edm jedom. Zistilo sa, \u017ee sa z\u00edskali r\u00f4zne v\u00fdsledky pre r\u00f4zne trvanie lie\u010dby a koncentr\u00e1cie.<br>V rakovinov\u00fdch bunk\u00e1ch, ktor\u00e9 boli o\u0161etren\u00e9 v\u010del\u00edm jedom s d\u00e1vkou 3,0 ug\/ml po\u010das 48 hod\u00edn, sa v porovnan\u00ed s kontroln\u00fdmi bunkami zaznamenal v\u00fdznamn\u00fd pokles Ramanov\u00fdch vibr\u00e1ci\u00ed prote\u00ednov a DNA.<\/p>\n\n\n\n<p>Toto zistenie vysoko nazna\u010duje, \u017ee doch\u00e1dza k degrad\u00e1cii a denatur\u00e1cii fragment\u00e1cie DNA a prote\u00ednov. V\u00fdsledky Ramanovej spektroskopie boli testom Western blot a TUNEL. Ramanova spektroskopia spolu s PCA teda vedie k neinvaz\u00edvnemu n\u00e1stroju na hodnotenie bunkov\u00fdch zmien prostredn\u00edctvom protirakovinov\u00fdch vlastnost\u00ed v\u010delieho jedu.<\/p>\n\n\n\n<p><strong>\u00davod<\/strong><br>Na dosiahnutie \u00fa\u010dinnej a bezpe\u010dnej lie\u010dby rakoviny je d\u00f4le\u017eit\u00e9 pochopi\u0165 interakciu r\u00f4znych protirakovinov\u00fdch medi\u00e1torov s postihnut\u00fdmi bunkami. Je tie\u017e dobre zn\u00e1me, \u017ee protirakovinov\u00e1 terapia vedie k apopt\u00f3ze cie\u013eov\u00fdch rakovinov\u00fdch buniek. Vy\u017eaduj\u00fa tie\u017e ve\u013ek\u00e9 mno\u017estvo materi\u00e1lu na z\u00edskanie len mal\u00e9ho mno\u017estva kone\u010dn\u00e9ho produktu. Okrem toho molekul\u00e1rny mechanizmus t\u00fdchto interakci\u00ed nie je mo\u017en\u00e9 hodnoti\u0165 prostredn\u00edctvom t\u00fdchto testov, preto\u017ee fluorescen\u010dn\u00e9 zna\u010dky pou\u017e\u00edvan\u00e9 po\u010das merania m\u00f4\u017eu vies\u0165 k zmen\u00e1m v biologick\u00fdch podmienkach. Na monitorovanie \u017eiv\u00fdch buniek v re\u00e1lnom \u010dase je teda potrebn\u00e1 neinvaz\u00edvna analytick\u00e1 technika bez ozna\u010denia.<\/p>\n\n\n\n<p>Technika pou\u017eit\u00e1 v tejto klinickej \u0161t\u00fadii \u2013 Ramanova spektroskopia \u2013 je r\u00fdchla a neinvaz\u00edvna technika, ktor\u00e1 dokonca nevy\u017eaduje \u017eiadne odbery vzoriek pred vykonan\u00edm anal\u00fdzy. Poskytuje tie\u017e podrobn\u00e9 kvantitat\u00edvne inform\u00e1cie o chemickom zlo\u017een\u00ed, molekul\u00e1rnej \u0161trukt\u00fare a interakcii v bunk\u00e1ch prostredn\u00edctvom vysokej selektivity a citlivosti. Ramanova spektroskopia sa pou\u017eila v r\u00f4znych klinick\u00fdch \u0161t\u00fadi\u00e1ch na stanovenie: \u00fa\u010dinkov aktinomyc\u00ednu D na l\u00edniu rakoviny p\u013e\u00fac a na meranie molekul\u00e1rnych zmien (\u010dasovo z\u00e1visl\u00fdch) v pridru\u017een\u00fdch bunk\u00e1ch po\u010das apopt\u00f3zy. Ale v tejto klinickej \u0161t\u00fadii sa Ramanova spektroskopia pou\u017e\u00edva po prv\u00fdkr\u00e1t na ur\u010denie protirakovinov\u00e9ho \u00fa\u010dinku v\u010delieho jedu na bunky rakoviny prsn\u00edka.<\/p>\n\n\n\n<p><strong>Met\u00f3da<\/strong><br>\u013dudsk\u00e9 bunky rakoviny prsn\u00edka boli z\u00edskan\u00e9 z American Type Culture Collection, USA. Bunky boli kultivovan\u00e9 v Dulbeccovom vylep\u0161enom Eagle m\u00e9diu, ktor\u00e9 bolo doplnen\u00e9 antibiotikami a 10% FBS (fet\u00e1lne bovinn\u00e9 s\u00e9rum), vo vlhkej atmosf\u00e9re s 95% vzduchu a 5% CO2. PBML (mononukle\u00e1rne lymfocyty \u013eudskej perif\u00e9rnej krvi) boli nesk\u00f4r izolovan\u00e9 z krvi z\u00edskanej od darcov pomocou gradientovej centrifug\u00e1cie Ficoll-hypaque.<\/p>\n\n\n\n<p>Z\u00edskan\u00e9 PBML boli rozdelen\u00e9 do 2 r\u00f4znych zna\u010diek: horn\u00fd p\u00e1s mal PBMC, zatia\u013e \u010do spodn\u00fd p\u00e1s mal neutrofily. PBML sa najsk\u00f4r zozbierali a potom sa trikr\u00e1t premyli v PBS. Bunky boli vystaven\u00e9 v\u010deliemu jedu so zlo\u017een\u00edm: 50 % melitt\u00edn, 2,5 % apam\u00edn, 2 % MCD peptid, 2 % am\u00edn, 1,5 % hyaluronid\u00e1za, 0,5 % histid\u00edn, 1 % lyso-PLA, 12 % PLA a 10 % a\u017e 17 % norepinefr\u00ednu, dopam\u00ednu, kyslej fosfomonoester\u00e1zy, invert\u00e1zy, glukozid\u00e1zy a inhib\u00edtora prote\u00e1zy \u2013 tieto zlo\u017eky boli takmer 100% \u010dist\u00e9 a boli inkubovan\u00e9 po dobu 24 hod\u00edn.<\/p>\n\n\n\n<p>Bunky boli nasaden\u00e9 na 96-jamkov\u00e9 platne a nesk\u00f4r o\u0161etren\u00e9 r\u00f4znymi d\u00e1vkami v\u010delieho jedu a fyziologick\u00e9ho roztoku po\u010das r\u00f4znych obdob\u00ed. Po ukon\u010den\u00ed lie\u010dby sa bunkov\u00e1 \u017eivotaschopnos\u0165 sk\u00famala pomocou s\u00fapravy Cell Counting Kit-8. Syst\u00e9m detekcie apopt\u00f3zy bol pou\u017eit\u00fd pod\u013ea pokynov dod\u00e1vate\u013ea. O\u0161etren\u00e9 bunky sa dvakr\u00e1t premyli PBC a potom sa na 20 min\u00fat pridal aj 1 % paraformaldehyd (\u013eadovo studen\u00fd). Bunky sa inkubovali jednu hodinu pri 37 stup\u0148och Celzia v tmavom prostred\u00ed v 50 ul tlmiv\u00e9ho roztoku pozost\u00e1vaj\u00faceho z fluoresce\u00ednu-12-dUTP, ktor\u00fd m\u00e1 tie\u017e koncov\u00fa deoxynukleotidyltransfer\u00e1zu na ozna\u010denie 3&#8242;-OH koncov fragmentovanej DNA.<\/p>\n\n\n\n<p>Po tejto reakcii boli bunky op\u00e4\u0165 premyt\u00e9 roztokom PBS a zafarben\u00e9 zmesou 250 ug RN\u00e1zy a 5,0 ug\/ml BSA. Povrchov\u00e1 expoz\u00edcia fosfatidylser\u00ednu apoptotick\u00fdmi bunkami bola hodnoten\u00e1 prietokov\u00fdm cytometrom po pridan\u00ed anex\u00ednu V-V-FITC a s\u00fa\u010dasne zafarben\u00e1 PI. Z\u00edskan\u00e9 vzorky sa sk\u00famali na prietokovom cytometri FACSort pomocou softv\u00e9ru Lysis II.<\/p>\n\n\n\n<p>Na uskuto\u010dnenie Ramanovho spektroskopick\u00e9ho merania sa pou\u017eil substr\u00e1t potiahnut\u00fd zlatom. Bola pridan\u00e1 vrstva Cr, aby mohla by\u0165 spr\u00e1vna adh\u00e9zia medzi sklenen\u00fdm substr\u00e1tom a zlat\u00fdm filmom. Z ka\u017edej skupiny sa odobralo 10 individu\u00e1lnych buniek. Ramanove spektr\u00e1 boli z\u00edskan\u00e9 pomocou konfok\u00e1lneho Ramanovho syst\u00e9mu SENTERRA vybaven\u00e9ho di\u00f3dov\u00fdm laserov\u00fdm zdrojom (785 nm a 100 mW) s rozl\u00ed\u0161en\u00edm 3 cm. Ramanove sign\u00e1ly sa zbierali cez 100x MPLN N.A 0,9 Olympus, ktor\u00fd generuje laserov\u00fd bod 1 um.<br>Predspracovanie \u00fadajov a z\u00edskavanie Ramanovho spektra bolo riaden\u00e9 softv\u00e9rom OPUS. Na extrakciu \u010distej vzorky Ramanov\u00fdch spektier sa na nameran\u00e9 \u00fadaje aplikoval automatizovan\u00fd algoritmus na odstr\u00e1nenie autofluorescen\u010dn\u00e9ho pozadia. Ramanove merania boli zaznamenan\u00e9 v akumula\u010dnom \u010dase 60 sek\u00fand v rozsahu 600 cm-1 a\u017e 1750 cm-1. Ramanove spektr\u00e1 vybran\u00fdch buniek sa vypo\u010d\u00edtali ako priemer z desiatich vzoriek.<\/p>\n\n\n\n<p><strong>V\u00fdsledky<\/strong><br>Na stanovenie cytotoxick\u00fdch \u00fa\u010dinkov v\u010delieho jedu na \u013eudsk\u00e9 norm\u00e1lne bunky a bunky rakoviny prsn\u00edka sa stanovila \u017eivotaschopnos\u0165 buniek pomocou testu CCK-8. Zistilo sa, \u017ee BV v\u00fdznamne br\u00e1ni prolifer\u00e1cii rakovinov\u00fdch buniek. Na \u010fal\u0161ie rozl\u00ed\u0161enie medzi n\u00e1chylnos\u0165ou norm\u00e1lnych buniek a rakovinov\u00fdch buniek sa sk\u00famali \u00fa\u010dinky v\u010delieho jedu v PBML. V\u00fdsledkom bolo, \u017ee BV nevykazoval \u017eiadny druh cytotoxick\u00e9ho \u00fa\u010dinku na PBLM v d\u00e1vke 12,4 ug\/ml po 72 hodin\u00e1ch inkub\u00e1cie.<\/p>\n\n\n\n<p>Zatia\u013e \u010do v\u00fdsledky z\u00edskan\u00e9 prostredn\u00edctvom Ramanovej spektroskopie v kombin\u00e1cii s PCA preuk\u00e1zali zhodu s v\u00fdsledkami z\u00edskan\u00fdmi prostredn\u00edctvom konven\u010dn\u00fdch biologick\u00fdch testov, ako s\u00fa testy western blot, TUNEL a \u017eivotaschopnos\u0165. T\u00e1to technika je najlep\u0161ia na ur\u010denie apoptotick\u00fdch \u00fa\u010dinkov v\u010delieho jedu prostredn\u00edctvom neinvaz\u00edvneho hodnotenia kvality bunkov\u00fdch zmien bez ozna\u010denia.<\/p>\n\n\n\n<p>V\u00fdsledky z\u00edskan\u00e9 z Ramanovej spektroskopie v kombin\u00e1cii s PCA uk\u00e1zali dobr\u00fa zhodu s v\u00fdsledkami z\u00edskan\u00fdmi pou\u017eit\u00edm konven\u010dn\u00fdch biologick\u00fdch testov, ako s\u00fa testy \u017eivotaschopnosti, TUNEL a western blot, ako je op\u00edsan\u00e9 v predch\u00e1dzaj\u00facej \u010dasti. Preto je k\u013e\u00fa\u010dov\u00e9 vyvin\u00fa\u0165 alternat\u00edvnu terapiu, ktor\u00e1 by mohla kontrolova\u0165 TNBC.
