Napriek tomu je potrebn\u00e9 intenz\u00edvne \u0161tudova\u0165 z\u00e1kladn\u00e9 mechanizmy hlavn\u00fdch zlo\u017eiek v\u010delieho jedu, ktor\u00e9 vyvol\u00e1vaj\u00fa protiples\u0148ov\u00e9 \u00fa\u010dinky.<\/p>\n\n\n\n<p>V tejto klinickej \u0161t\u00fadii sa melit\u00edn a apam\u00edn, ktor\u00e9 s\u00fa zn\u00e1me svojimi protiz\u00e1palov\u00fdmi \u00fa\u010dinkami, sk\u00famali v surovej forme a produkte na b\u00e1ze hmly, oddelene na kol\u00f3ni\u00e1ch T. rubrum, aby sa ur\u010dili ich protiples\u0148ov\u00e9 \u00fa\u010dinky.<\/p>\n\n\n\n<p><strong>Met\u00f3da<\/strong><br>Kol\u00f3nie Apis mellifera boli starostlivo udr\u017eiavan\u00e9 v N\u00e1rodnej akad\u00e9mii po\u013enohospod\u00e1rskej vedy v K\u00f3rei. V\u010del\u00ed jed bol zozbieran\u00fd pomocou zbern\u00e9ho zariadenia od Chung Jin Biotech Co. Korea. Hlavn\u00fdm cie\u013eom bolo prin\u00fati\u0165 v\u010dely bodn\u00fa\u0165 do sklenenej dosky a to sa dosiahlo vysok\u00fdm elektrick\u00fdm pr\u00fadom smeruj\u00facim do \u00fa\u013ea. Jed sa nechal uschn\u00fa\u0165 a nesk\u00f4r sa zo\u0161krabal a zhroma\u017edil na jednom mieste. V\u010del\u00ed jed sa potom zriedil v sterilnej vode, uistil sa, \u017ee voda je dostato\u010dne studen\u00e1, a potom sa odstre\u010foval pri 10 000 g takmer 5 min\u00fat pri teplote 4 stupne Celzia. Tento proces sa uskuto\u010dnil na odstr\u00e1nenie ak\u00e9hoko\u013evek druhu zvy\u0161ku zo supernatantu.<\/p>\n\n\n\n<p>Po tomto procese bol v\u010del\u00ed jed lyofilizovan\u00fd v lyofiliza\u010dnej su\u0161i\u010dke a potom op\u00e4\u0165 schladen\u00fd na 4 stupne Celzia. V\u010del\u00ed jed pou\u017eit\u00fd v tejto klinickej \u0161t\u00fadii bol overen\u00fd g\u00e9lovou chromatografiou s vylu\u010dovacou ve\u013ekos\u0165ou vykonanou jeho rozpusten\u00edm v 0,02 M fosf\u00e1tovom pufri spolu s 0,25 M NaCl s pH 7,2. \u010eal\u0161ie zlo\u017eky vr\u00e1tane apam\u00ednu a melit\u00ednu boli z\u00edskan\u00e9 od Sigma, USA a BBM a Dongsung Pharmaceuticals, K\u00f3rea.<\/p>\n\n\n\n<p>Antifung\u00e1lny mechanizmus bol stanoven\u00fd sk\u00faman\u00edm zmien plochy vykonan\u00fdch v ka\u017edej skupine, ktor\u00e1 bola predmetom \u0161t\u00fadie po\u010das 14 dn\u00ed \u2013 v intervale dvoch. Pozorovalo sa, \u017ee experiment\u00e1lne skupiny o\u0161etren\u00e9 surov\u00fdm v\u010del\u00edm jedom mali pomal\u00fd rastov\u00fd rozdiel v porovnan\u00ed s druhou skupinou (kontroln\u00e1 skupina) pri koncentr\u00e1ci\u00e1ch 10 mg\/100 ul a 40 mg\/100 ul. Pozoruhodn\u00fd rozdiel v r\u00fdchlosti rastu skup\u00edn o\u0161etren\u00fdch BBM medzi experiment\u00e1lnymi skupinami pri koncentr\u00e1ci\u00e1ch 200 ul a 300 ul bol pozorovan\u00fd po\u010das celkovej kontroly.<\/p>\n\n\n\n<p><strong>Diskusia<\/strong><br>Hlavn\u00fdm cie\u013eom tejto klinickej \u0161t\u00fadie bolo pos\u00fadi\u0165 mechanizmus zlo\u017eiek v\u010delieho jedu, ktor\u00e9 zastavuj\u00fa prejav ples\u0148ov\u00e9ho patog\u00e9nu T. rubrum. V tejto klinickej \u0161t\u00fadii sa porovn\u00e1vali antifung\u00e1lne \u00fa\u010dinky medzi kontroln\u00fdmi skupinami a experiment\u00e1lnymi skupinami v \u0161pecifick\u00fdch \u010dasov\u00fdch intervaloch, iba v\u010del\u00ed jed v surovej forme a produkt na b\u00e1ze hmly vykazovali \u0161tatisticky v\u00fdznamn\u00e9 hodnoty.<\/p>\n\n\n\n<p>Pri \u010fal\u0161om spracovan\u00ed, \u010dasov\u00fd bod predstavuj\u00faci protiples\u0148ov\u00fd \u00fa\u010dinok RBV bol z\u00e1visl\u00fd od d\u00e1vky, preto\u017ee 40 mg\/100 ul (p=0,000) a 10 mg\/100 ul (p=0,026) koncentrovan\u00e9 vzorky vykazovali \u0161tatisticky v\u00fdznamn\u00fd rozdiel v ploche v porovnan\u00ed s 0,1 mg \/100 ul (p = 0,084). Anal\u00fdza uskuto\u010dnen\u00e1 medzi kontroln\u00fdmi skupinami a experiment\u00e1lnymi skupinami nepredstavovala \u017eiadny druh \u0161tatistick\u00e9ho rozdielu s\u00favisiaceho s rastom h\u00fab na akejko\u013evek \u00farovni koncentr\u00e1cie. Rovnak\u00e9 v\u00fdsledky boli zobrazen\u00e9 v kol\u00f3ni\u00e1ch, ktor\u00e9 boli o\u0161etren\u00e9 apam\u00ednom. Zatia\u013e \u010do zostupn\u00e1 reakcia zabra\u0148uje transkripcii z\u00e1palov\u00e9ho cytok\u00ednu, ktor\u00fd m\u00e1 nieko\u013eko ochrann\u00fdch \u00fa\u010dinkov indukovan\u00fdch melitt\u00ednom.<\/p>\n\n\n\n<p>Z\u00e1ver T\u00e1to klinick\u00e1 \u0161t\u00fadia prezentovala antifung\u00e1lne \u00fa\u010dinky, ktor\u00e9 maj\u00fa zlo\u017eky BV spolu s kozmetick\u00fdm produktom zalo\u017een\u00fdm na BV, ale s\u00fa potrebn\u00e9 \u010fal\u0161ie experimenty, ktor\u00e9 maj\u00fa ve\u013ek\u00fa z\u00e1klad\u0148u vzoriek a viacer\u00e9 druhy h\u00fab a r\u00f4zne zlo\u017eky F BV. Albicans. U niektor\u00fdch pacientov sa m\u00f4\u017ee diseminovan\u00e1 kandid\u00f3za zmeni\u0165 na \u017eivot ohrozuj\u00face ochorenie. Tento typ funkcie v\u00e4\u010d\u0161inou vedie k infekci\u00e1m, ako je drozd a vaginit\u00edda. Amfoteric\u00edn B sa be\u017ene predpisuje na lie\u010dbu ples\u0148ovej infekcie, zatia\u013e \u010do azoly sa pou\u017e\u00edvaj\u00fa v r\u00f4znych klinick\u00fdch situ\u00e1ci\u00e1ch. Na prekonanie t\u00fdchto probl\u00e9mov s\u00fa potrebn\u00e9 prirodzen\u00e9 terapeutick\u00e9 opatrenia.<\/p>\n\n\n\n<p>T\u00e1to klinick\u00e1 \u0161t\u00fadia bola vykonan\u00e1 s cie\u013eom presk\u00fama\u0165 antifung\u00e1lne vlastnosti SBV a BV proti C. albicans izolovan\u00fdm z\u00edskan\u00fdm z vag\u00edny a vzoriek krvi.<\/p>\n\n\n\n<p><strong>Met\u00f3da<\/strong><br>SBV a BV boli vytvoren\u00e9 v K\u00f3rejskom farmakopunkt\u00farnom in\u0161tit\u00fate v K\u00f3rei. Lyofilizovan\u00e9 cel\u00e9 SBV a BV sa zmie\u0161ali v destilovanej vode. Pou\u017eili sa r\u00f4zne koncentr\u00e1cie tejto zmesi. Na sk\u00famanie antifung\u00e1lnych vlastnost\u00ed SBV a BV bol C. Albicans pou\u017eit\u00fd \u0161tandardn\u00fd kme\u0148.
