V kone\u010dnom d\u00f4sledku sp\u00f4sobuje paral\u00fdzu a stratu svalovej hmoty. V nieko\u013ek\u00fdch pr\u00edpadoch bolo hl\u00e1sen\u00e9, \u017ee t\u00e1to choroba sa \u0161\u00edri na nohy, ruky a dokonca aj na jazyk.<\/p>\n\n\n\n<p>V\u00fdskumn\u00edci na\u0161li r\u00f4zne te\u00f3rie, ktor\u00e9 pom\u00e1haj\u00fa vysvetli\u0165 d\u00f4vody prejavu ALS. Predpoklad\u00e1 sa, \u017ee \u0161truktur\u00e1lne zmeny prote\u00ednov m\u00f4\u017eu vies\u0165 k poruche a smrti nervov\u00fdch buniek. V oblasti ALS sa za posledn\u00fdch 10 rokov vykonal zna\u010dn\u00fd v\u00fdskum. Jedn\u00fdm z najv\u00fdznamnej\u0161\u00edch je eur\u00f3psky projekt, ktor\u00fd pozost\u00e1va z 15 000 subjektov ALS. S\u00fa\u010das\u0165ou tohto rozsiahleho projektu sa stane aj v\u00fdskumn\u00e1 skupina z Umea University, ktor\u00e1 m\u00e1 500 \u0161v\u00e9dskych pacientov s ALS.<\/p>\n\n\n\n<p>Najbe\u017enej\u0161\u00ed typ ALS pozost\u00e1va z SOD1, chybne poskladan\u00e9ho prote\u00ednu, ktor\u00fd sa hromad\u00ed vo vysiela\u010doch neur\u00f3nov. V\u00fdskumn\u00edci sa zameriavaj\u00fa na d\u00f4vody tohto nespr\u00e1vneho poskladania a ako sa \u0161\u00edri a ovplyv\u0148uje nervov\u00fd syst\u00e9m. Hlavn\u00fdm zameran\u00edm r\u00f4znych v\u00fdskumn\u00fdch skup\u00edn je navrhn\u00fa\u0165 met\u00f3dy, ktor\u00e9 m\u00f4\u017eu pom\u00f4c\u0165 zabr\u00e1ni\u0165 tvorbe a produkcii SOD1. Je potrebn\u00e9 poznamena\u0165, \u017ee nieko\u013eko typov ALS a FTD (demencia kotlov\u00e9ho laloku) m\u00e1 takmer rovnak\u00fd chorobn\u00fd proces, ktor\u00fd postihuje r\u00f4zne \u010dasti nervov\u00e9ho syst\u00e9mu. Pod\u013ea vedcov je to sp\u00f4soben\u00e9 t\u00fdm, \u017ee obe choroby maj\u00fa rovnak\u00e9 g\u00e9ny. St\u00e1le je potrebn\u00e9 pos\u00fadi\u0165, pre\u010do sa niektor\u00ed pacienti stan\u00fa obe\u0165ou FTD a in\u00ed ALS.<\/p>\n\n\n\n<p><strong>Melitt\u00edn obnovuje funkciu proteaz\u00f3mu vo zvieracom modeli ALS<\/strong><br>Amyotrofick\u00e1 later\u00e1lna skler\u00f3za, tie\u017e zn\u00e1ma ako ALS, je chronick\u00e1 paralyzuj\u00faca porucha, ktor\u00e1 je sp\u00f4soben\u00e1 zhor\u0161en\u00edm motorick\u00fdch neur\u00f3nov\/ m\u00f4\u017ee sa vyskytn\u00fa\u0165 ako famili\u00e1rne, tak aj sporadick\u00e9 ochorenie. MtSOD1 (mutantn\u00fd SOD1) pr\u00edtomn\u00fd v motorick\u00fdch neur\u00f3noch sa st\u00e1va zranite\u013en\u00fdm vo\u010di tomuto ochoreniu v d\u00f4sledku nespr\u00e1vneho poskladania prote\u00ednov, neuroz\u00e1palu, excitotoxicity, defektnej signaliz\u00e1cie rastov\u00e9ho faktora a axon\u00e1lneho transportu, cytoskelet\u00e1lnych abnormal\u00edt, oxida\u010dn\u00e9ho po\u0161kodenia a mitochondri\u00e1lnej dysfunkcie.<\/p>\n\n\n\n<p>V\u010del\u00ed jed je nabit\u00fd z\u00e1kladn\u00fdmi lie\u010div\u00fdmi zlo\u017ekami. Melitt\u00edn je jedn\u00fdm z t\u00fdch prvkov, ktor\u00e9 sa nach\u00e1dzaj\u00fa vo v\u010delom jede. Je to re\u0165azec 26 aminokysel\u00edn, ktor\u00fd sa u\u017e dlho pou\u017e\u00edva v r\u00f4znych tradi\u010dn\u00fdch \u010d\u00ednskych liekoch na \u00fa\u013eavu od bolesti a z\u00e1palu. Hlavn\u00fdm \u00fa\u010delom tejto \u0161t\u00fadie bolo pos\u00fadi\u0165 lie\u010div\u00e9 pr\u00ednosy melitt\u00ednu u subjektov s ALS. Pre t\u00fato klinick\u00fa \u0161t\u00fadiu bola vybran\u00e1 be\u017ene pou\u017e\u00edvan\u00e1 my\u0161 hSOD1G93A, ktor\u00e1 zdedila ALS. Melitt\u00edn bol podan\u00fd v akupunkt\u00farnom bode ST36, aby sa zistilo, \u010di m\u00f4\u017ee pom\u00f4c\u0165 zabr\u00e1ni\u0165 nespr\u00e1vnemu poskladaniu prote\u00ednov a potla\u010di\u0165 stratu neur\u00f3nov.<\/p>\n\n\n\n<p>Zistilo sa, \u017ee my\u0161 hSOD1G93A lie\u010den\u00e1 melit\u00ednom vykazovala jednozna\u010dn\u00e9 zn\u00ed\u017eenie po\u010dtu mikrogli\u00ed a fosfo-p38 v mozgovom pr\u00fade a mieche. Melittin tie\u017e pomohol zlep\u0161i\u0165 stav neur\u00f3nov v mieche a celkov\u00fa motorick\u00fa funkciu v porovnan\u00ed s kontrolnou skupinou. V\u00fdrazn\u00e9 zv\u00fd\u0161enie modifik\u00e1cie a-synukle\u00ednu, ako je nitr\u00e1cia a fosforyl\u00e1cia, bolo tie\u017e pozorovan\u00e9 v mieche a hlavnom mozgovom kmeni my\u0161i hSOD1G93A.<\/p>\n\n\n\n<p>Nakoniec mo\u017eno poveda\u0165, \u017ee pod\u00e1vanie melitt\u00ednu zn\u00ed\u017eilo chybn\u00e9 poskladanie a-synukle\u00ednu a obnovilo proteazom\u00e1lnu aktivitu v mieche a mozgovom kmeni symptomatick\u00fdch my\u0161\u00ed. V\u00fdskum nazna\u010duje, \u017ee existuje potenci\u00e1lna s\u00favislos\u0165 medzi pod\u00e1van\u00edm melitt\u00ednu a retrogresiou neuroz\u00e1palu u my\u0161\u00edch modelov ALS hSOD1G93A.<\/p>\n\n\n\n<p><strong>V\u010del\u00ed jed zmier\u0148uje neuroz\u00e1palov\u00e9 udalosti a predl\u017euje \u017eivot<\/strong><br>ALS znamen\u00e1 amyotrofick\u00fa later\u00e1lnu skler\u00f3zu. Ide o ochorenie, ktor\u00e9 postihuje centr\u00e1lny nervov\u00fd syst\u00e9m a sp\u00f4sobuje r\u00f4zne poruchy. Odumieranie motorick\u00fdch neur\u00f3nov je hlavnou pr\u00ed\u010dinou tohto mno\u017eiaceho sa ochorenia \u2013 m\u00f4\u017ee by\u0165 famili\u00e1rneho p\u00f4vodu alebo sporadick\u00e9. Genetick\u00fd faktor, ktor\u00fd prispieva k vzniku ALS, je mutant SOD1, ktor\u00fd vyvol\u00e1va zranite\u013enos\u0165 motorick\u00fdch neur\u00f3nov v d\u00f4sledku nespr\u00e1vneho poskladania prote\u00ednov, neuroz\u00e1palu, glutam\u00e1tovej excitotoxicity, defektn\u00e9ho axon\u00e1lneho transportu, cytoskelet\u00e1lnych abnormal\u00edt, oxida\u010dn\u00e9ho po\u0161kodenia a mitochondri\u00e1lnej dysfunkcie.<\/p>\n\n\n\n<p>BV (v\u010del\u00ed jed) sa v orient\u00e1lnej medic\u00edne pou\u017e\u00edva u\u017e dlho. N\u00e1le\u017eit\u00e9 liter\u00e1rne d\u00f4kazy nazna\u010duj\u00fa, \u017ee v\u010del\u00ed jed m\u00e1 protiz\u00e1palov\u00e9 vlastnosti proti z\u00e1palov\u00fdm reakci\u00e1m, ktor\u00e9 s\u00fa spojen\u00e9 s r\u00f4znymi z\u00e1palov\u00fdmi ochoreniami, ako je artrit\u00edda. Hlavn\u00fdm bodom vykonanej \u0161t\u00fadie bolo analyzova\u0165 \u00fa\u010dinok v\u010delieho jedu na mutantn\u00e9 my\u0161i hSOD1G93A, \u010di BV \/v\u010del\u00ed jed\/ pom\u00e1ha pri potl\u00e1\u010dan\u00ed straty motorick\u00fdch neur\u00f3nov alebo nie.<\/p>\n\n\n\n<p>BV (v\u010del\u00ed jed) sa bilater\u00e1lne pod\u00e1val v ST36 acupoint dvojt\u00fd\u017ed\u0148ov\u00fdm samcom my\u0161\u00ed hSOD1G93A. Zusanli (ST36) acupoint je vybran\u00fd, preto\u017ee je zn\u00e1me, \u017ee sprostredk\u00fava protiz\u00e1palov\u00fd \u00fa\u010dinok. Na kontrolu motorickej aktivity bola vykonan\u00e1 \u0161peci\u00e1lna anal\u00fdza naz\u00fdvan\u00e1 rotarod test. \u0160tatistika pre\u017eitia bola sk\u00faman\u00e1 pomocou met\u00f3dy Kaplan-Meierovej krivky pre\u017eitia. Celkov\u00fd \u00fa\u010dinok lie\u010dby v\u010del\u00edm jedom na protineuroz\u00e1pal samcov my\u0161\u00ed hSOD1G93A sa hodnotil prostredn\u00edctvom imunoreakci\u00ed s pou\u017eit\u00edm protil\u00e1tky TNF \u2013 \u03b1 a Iba 1 ako markera mikroglie. Aktiv\u00e1cia prote\u00ednov kasp\u00e1zy 3, p 38 MAP kin\u00e1za a ERK sa zv\u00e1\u017eili pou\u017eit\u00edm Western blottingu.<\/p>\n\n\n\n<p>Transg\u00e9nne my\u0161i HSOD1G93A, ktor\u00e9 boli lie\u010den\u00e9 v\u010del\u00edm jedom, vykazovali v\u00fdznamn\u00fd pokles hladiny fosforu-p38 MAPK a markera mikroglie v mozgovom kmeni a mieche. Okrem toho pod\u00e1vanie v\u010delieho jedu subjektom so symptomatickou ALS pomohlo zlep\u0161i\u0165 celkov\u00fa motorick\u00fa aktivitu. Medi\u00e1n pre\u017eitia v porovnan\u00ed s kontrolnou skupinou zvierat lie\u010den\u00fdch v\u010del\u00edm jedom bol o 18 % vy\u0161\u0161\u00ed. Zistilo sa tie\u017e, \u017ee v\u010del\u00ed jed pom\u00e1hal pri blokovan\u00ed nedostatkov morfol\u00f3gie kr\u00eds a mitochondri\u00e1lnej \u0161trukt\u00fary a potl\u00e1\u010dal hladiny aktivity kasp\u00e1zy-3 v mieche my\u0161\u00ed s ALS.<\/p>\n\n\n\n<p>Vo svetle tejto klinickej \u0161t\u00fadie v\u00fdskum nazna\u010duje, \u017ee v\u010del\u00ed jed mo\u017eno pou\u017ei\u0165 ako \u00fa\u010dinn\u00fd terapeutick\u00fd liek na rie\u0161enie \u00fa\u010dinkov ALS u mutantn\u00fdch my\u0161\u00ed hSOD1G93A.<\/p>\n\n\n\n<p><strong>Melitt\u00edn obnovuje funkciu proteaz\u00f3mu vo zvieracom modeli ALS<\/strong><br>ALS je jedn\u00fdm z najr\u00fdchlej\u0161ie progreduj\u00facich neurodegenerat\u00edvnych ochoren\u00ed, ktor\u00e9 je sp\u00f4soben\u00e9 smr\u0165ou neur\u00f3nov v horn\u00fdch motorick\u00fdch neur\u00f3noch a mieche. V d\u00f4sledku toho vedie k paral\u00fdze pripojen\u00fdch svalov. Transmut\u00e1cie v SOD1 (Zn\/Cu superoxiddismut\u00e1za) predstavuj\u00fa takmer 20 % dedi\u010dn\u00fdch pr\u00edpadov FALS (ALS). U transg\u00e9nnych my\u0161\u00ed hSOD1G93A sa vyvinuli kardin\u00e1lne sympt\u00f3my amyotrofickej later\u00e1lnej skler\u00f3zy u \u013eud\u00ed, ktor\u00e9 zah\u0155\u0148aj\u00fa svalov\u00fa atrofiu a paral\u00fdzu. Patofyziologick\u00e9 mechanizmy amyotrofickej later\u00e1lnej skler\u00f3zy zah\u0155\u0148aj\u00fa z\u00e1pal, naru\u0161en\u00fd axon\u00e1lny transport, glutam\u00e1tov\u00fa excitotoxicitu a mitochondri\u00e1lnu dysfunkciu.<\/p>\n\n\n\n<p>V\u010del\u00ed jed sa z\u00edskava priamo z v\u010diel medonosn\u00fdch a jeho v\u0161eobecn\u00fd n\u00e1zov je apitox\u00edn. Tradi\u010dne sa pou\u017e\u00edva na kontrolu z\u00e1palu u subjektov s osteoartrit\u00eddou a chronickou reumatoidnou artrit\u00eddou. V predch\u00e1dzaj\u00facich v\u00fdskumoch sa u\u017e zistilo, \u017ee v\u010del\u00ed jed m\u00e1 protineuroz\u00e1palov\u00e9 vlastnosti. V\u010del\u00ed jed m\u00e1 ve\u013ea peptidov, ako je apam\u00edn a melitt\u00edn a \u010fal\u0161ie bioakt\u00edvne zl\u00fa\u010deniny. Melitt\u00edn je siln\u00fd aminokyselinov\u00fd polypeptid, ktor\u00fd tvor\u00ed takmer 60 % such\u00e9ho v\u010delieho jedu. Tie\u017e sa predpoklad\u00e1, \u017ee je to popredn\u00e1 biologicky akt\u00edvna l\u00e1tka nach\u00e1dzaj\u00faca sa vo v\u010delom jede, ktor\u00e1 pri registr\u00e1cii na akupunkt\u00fare generuje protiz\u00e1palov\u00e9 a antinocicept\u00edvne \u00fa\u010dinky.<\/p>\n\n\n\n<p>My\u0161i HSOD1G93A sa pou\u017eili na laborat\u00f3rnu \u0161t\u00fadiu pod\u013ea usmernen\u00ed stanoven\u00fdch N\u00e1rodn\u00fdm in\u0161tit\u00fatom zdravia (Bethesda, MD). Z\u00e1kladn\u00e9 protokoly ods\u00fahlasili v\u00fdbory pre pou\u017e\u00edvanie K\u00f3rejsk\u00e9ho in\u0161tit\u00fatu orient\u00e1lnej medic\u00edny a in\u0161titucion\u00e1lnej starostlivosti o zvierat\u00e1. Transg\u00e9nne my\u0161i B6SJL boli z\u00edskan\u00e9 z Jackson Laboratory \u2013 nes\u00face modifik\u00e1ciu glyc\u00ednu na alan\u00edn na 93. kod\u00f3ne cytosolick\u00e9ho genetick\u00e9ho typu Zn\/CU superoxiddismut\u00e1zy (HSOD1G93A). V\u0161etk\u00fdm subjektom (my\u0161i0 bol poskytnut\u00fd dostato\u010dn\u00fd pr\u00edstup k vode, potrave pre hlodavce a \u0161tandardn\u00e9mu ustajneniu).<\/p>\n\n\n\n<p>Z lie\u010debn\u00fdch d\u00f4vodov bol Melittin z\u00edskan\u00fd zo Sigma St. Louis, MO a nesk\u00f4r zrieden\u00fd trochou fyziologick\u00e9ho roztoku. My\u0161iam samcom vo veku 98 t\u00fd\u017ed\u0148ov bol bilater\u00e1lne injikovan\u00fd melit\u00edn v akupunkt\u00farnom bode ST36. Je zn\u00e1me, \u017ee tento bod m\u00e1 protiz\u00e1palov\u00e9 \u00fa\u010dinky. Samcom my\u0161\u00ed bol pod\u00e1van\u00fd melitt\u00edn dvakr\u00e1t t\u00fd\u017edenne. Zvierat\u00e1m z kontrolnej skupiny zodpovedaj\u00faceho veku sa obojstranne podali rovnak\u00e9 objemy fyziologick\u00e9ho roztoku v rovnakom akutnom bode.<\/p>\n\n\n\n<p>My\u0161i vo veku 98 t\u00fd\u017ed\u0148ov boli nesk\u00f4r rozdelen\u00e9 do dvoch r\u00f4znych skup\u00edn: my\u0161i lie\u010den\u00e9 fyziologick\u00fdm roztokom a my\u0161i lie\u010den\u00e9 melit\u00ednom. D\u013a\u017eka \u017eivota lie\u010den\u00fdch subjektov bola meran\u00e1 pomocou Kaplan Meierov\u00fdch kriviek pre\u017eitia pomocou softv\u00e9ru Sigmaplot 10 a softv\u00e9ru Prism 5.0. Z\u00edskan\u00e9 hodnoty boli starostlivo analyzovan\u00e9 jednosmern\u00fdm testom ANOVA a Dunnov\u00fdm viacn\u00e1sobn\u00fdm porovn\u00e1vac\u00edm testom.<\/p>\n\n\n\n<p>Zistilo sa, \u017ee spr\u00e1vny re\u017eim melitt\u00ednu pomohol zv\u00fd\u0161i\u0165 motorick\u00fa v\u00fdkonnos\u0165 u hSOD1G93A transg\u00e9nnych my\u0161\u00ed. Pod\u013ea v\u00fdsledkov behavior\u00e1lneho testu na rotarode sa motorick\u00e9 fungovanie zv\u00fd\u0161ilo 1,7-n\u00e1sobne po 7 d\u0148och lie\u010denia my\u0161\u00ed hSOD1G93A. My\u0161i HSOD1G93A lie\u010den\u00e9 10 dn\u00ed melit\u00ednom vykazovali p\u00f4sobiv\u00e9 2,8-n\u00e1sobn\u00e9 zlep\u0161enie motorickej aktivity.<\/p>\n\n\n\n<p>Tie\u017e sa zistilo, \u017ee paral\u00fdza a n\u00e1stup ochorenia sa oneskorili o 1 t\u00fd\u017ede\u0148 u my\u0161\u00ed, ktor\u00e9 boli lie\u010den\u00e9 melit\u00ednom, v porovnan\u00ed s t\u00fdmi, ktor\u00fdm bol pod\u00e1van\u00fd iba fyziologick\u00fd roztok. Zistilo sa v\u0161ak, \u017ee lie\u010dba melit\u00ednom neovplyv\u0148uje mieru pre\u017eitia alebo rozp\u00e4tie my\u0161\u00ed, bez oh\u013eadu na met\u00f3dy, ktor\u00e9 boli \u0161tudovan\u00e9 v laborat\u00f3riu. Medi\u00e1n pre\u017e\u00edvania kontrolnej skupiny bol trochu bl\u00edzky skupine lie\u010denej melit\u00ednom \u2013 129 a 132 dn\u00ed. Jedn\u00fdm z najpozoruhodnej\u0161\u00edch objavov bolo, \u017ee my\u0161i lie\u010den\u00e9 melit\u00ednom vykazovali oneskoren\u00fd n\u00e1stup ochorenia v porovnan\u00ed s my\u0161ami rovnak\u00e9ho veku, ktor\u00fdm bol pod\u00e1van\u00fd fyziologick\u00fd roztok.<\/p>\n\n\n\n<p>V\u00fdsledky teda ukazuj\u00fa, \u017ee lie\u010dba melit\u00ednom po\u010das ur\u010dit\u00e9ho obdobia pom\u00e1ha oddiali\u0165 progresiu ochorenia a vznik motorickej dysfunkcie, ale neprispieva k pred\u013a\u017eeniu \u017eivotnosti vybran\u00fdch my\u0161\u00ed.<\/p>\n\n\n\n<p>Laborat\u00f3rna \u0161t\u00fadia odha\u013euje, \u017ee melit\u00edn v\u00fdznamne zni\u017euje neuroz\u00e1palov\u00fd v\u00fdvoj a pom\u00e1ha zni\u017eova\u0165 modifik\u00e1ciu a-synukle\u00ednu zv\u00fd\u0161en\u00edm aktivity proteaz\u00f3mu u my\u0161\u00ed. Nazna\u010duje to, \u017ee chaper\u00f3nov\u00e9 prote\u00edny a proteaz\u00f3mov\u00e1 aktivita s\u00fa zodpovedn\u00e9 za odstr\u00e1nenie ak\u00fdchko\u013evek nespr\u00e1vne zlo\u017een\u00fdch prote\u00ednov, ako je synukle\u00edn a SOD1.<\/p>\n\n\n\n<p>Pridanie Okrem toho spojenie medzi PD a ALS zost\u00e1va nejasn\u00e9. Ke\u010f s\u00fa glutam\u00e1tov\u00e9 receptory naru\u0161en\u00e9, m\u00f4\u017ee to vies\u0165 k excitotoxicite. Nadmern\u00e1 stimul\u00e1cia NMDA receptorov prostredn\u00edctvom glutam\u00e1tu m\u00f4\u017ee vies\u0165 k abnorm\u00e1lnej neuron\u00e1lnej degener\u00e1cii, po ktorej nasleduj\u00fa z\u00e1chvaty, hypoglyk\u00e9mia, isch\u00e9mia a hypoxia. Z\u00edskan\u00e9 v\u00fdsledky m\u00f4\u017eu ma\u0165 ak\u00e9ko\u013evek klinick\u00e9 d\u00f4sledky a \u017ee BV mo\u017eno pou\u017ei\u0165 ako potenci\u00e1lnu lie\u010dbu na prevenciu neurogenerat\u00edvnych a z\u00e1palov\u00fdch ochoren\u00ed.<\/p>\n\n\n\n<p>V\u010del\u00ed jed, tween-20, L-glutam\u00e1t, 5-difenyltetraz\u00f3liumbromid a 3-(4,5-dimetyltiazol-2-yl) boli z\u00edskan\u00e9 od Sigma, USA. Zatia\u013e \u010do lakt\u00e1tdehydrogen\u00e1zov\u00e1 s\u00faprava bola z\u00edskan\u00e1 od Biovision Research Products, USA a fet\u00e1lne bovinn\u00e9 s\u00e9rum od Gibco. Prim\u00e1rne protil\u00e1tky potrebn\u00e9 pre p38, p-p38, ERK, p-ERK, AKT, JNK, p-JNK boli z\u00edskan\u00e9 od Cell Signaling Technology, USA.<br>Cytotoxicita bunkovej membr\u00e1ny bola sk\u00faman\u00e1 pomocou testu uvo\u013e\u0148ovania LDH, ktor\u00fd kvantitat\u00edvne spracov\u00e1va aktivitu LDH &#8211; stabiln\u00e9ho enz\u00fdmu, ktor\u00fd sa uvo\u013e\u0148uje, ke\u010f bunka dostane nejak\u00fd druh po\u0161kodenia.
